چکیده

Human peripheral blood lymphocytes can be phenotypically identified by the presence of one or both of two proteins. 225,000-dalton macromolecular insoluble cold globulin (225-MICG) and 185,000-dalton MICG (185-MICG). T cells synthesize and insert into their plasma membrane 225MICG. null cells 1 85-MICG. and B cells both 225 and 1 85-MICG. In contrast, the monoclonal B cells of chronic lymphocytic leukemia are characterized by the presence of 225-MICG and the absence of 1 85-MICG. We have recently found it possible to chemically deplete 1 85-MICG from viable normal B cells by treating them with diisopropylfluorophosphate (DFP). thus making normal B cells phenotypically resemble leukemic cells. In the present report we

Adenylate cyclase and guanylate cyclase activity in normal and leukemic human lymphocytes.

Adenylate cyclase (AC) and guanylate cyclase (GC) activities were studied in normal B-enriched and T-enriched lymphocytes, in lymphocytes of children with acute lymphocytic leukemia (ALL), and in lymphocytes of adults with chronic lymphocytic leukemia (CLL). AC activity was greater in normal B than T lymphocytes (215 pmole/min/mg protein versus 80 pmole in the membrane-enriched fraction) and i ...

Decrease and altered distribution of human T antigen on chronic lymphatic leukemia cells of T type, suggesting a clonal origin.

B- and T-cell markers were studied in a patient with chronic lymphocytic leukemia and erythroderma. The absence of immunoglobulin, complement receptor, and Fc receptor, and the presence of sheep erythrocyte receptor and T-cell antigen on the membrane of the leukemic cells classified them as thymus derived. Using quantitative microphotometric immunoautoradiography, surface antigen densities were...

Chronic lymphocytic leukemic B cells but not normal B cells are rescued from apoptosis by contact with normal bone marrow stromal cells.

The leukemic B lymphocytes from chronic lymphocytic leukemic (CLL) patients have a long survival in vivo, although ex vivo they rapidly die by apoptosis. To further investigate the mechanism of this, we have studied the influence of bone marrow stromal cells from normal subjects on apoptosis of B-CLL cells and normal umbilical cord blood (UCB) B lymphocytes. After 48 hours of incubation in medi...

Interferon-alpha alters spectrin organization in normal and leukemic human B lymphocytes.

Interferon-alpha (IFN-alpha) regulates the growth, differentiation, and recirculation of normal and malignant B lymphocytes. In this report we examine the effects of IFN-alpha on the distribution of the cytoskeletal protein spectrin in peripheral blood B lymphocytes from normal donors and patients diagnosed with chronic lymphocytic leukemia (CLL) and hairy cell leukemia (HCL). Exposure of norma...
